Interactions between mutations of p53 and K-ras genes in stomach cancer tissues, Helicobacter pylori infection, and IL-1B polymorphism

Abstract
Backgrounds: The aim of the study is to assess the effect of cigarette smoking, the IL-1B polymorphism, history of H. pylori infection on gastric cancer and to analyze the association between H. pylori infection and mutations of p53 and K-ras genes in gastric cancer tissue.

Methods: One hundred and ten gastric cancer patients and 220 age- and sex-matched controls were included in
this study. Information on the smoking habit was gathered, and the IL-1B genotype was identified for every subject. Mutations in the exons of p53 and K-ras genes were checked by sequencing. Relationships of cigarette smoking, IL-1B genotype, and anti-H. pylori IgG antibody with the risk of gastric cancer, and with mutations of the p53 and K-ras genes in gastric cancer tissue were statistically tested.

Results: The smoking prevalence was higher in cases than in controls, but cumulative smoking amount was
not. There was no significant difference in H. pylori infection rate and in IL-1B polymorphic distribution between cases and controls. Cigarette smoking and IL-1B polymorphism were not significantly associated with H. pylori infection. The mutation rates of p53 and K-ras genes in cancer tissue did not differ according to the smoking, IL-1B polymorphism or H. pylori infection in cases.

Conslusions: Smoking increases the risk of gastric cancer, but doest not that of H. pylori infection. H. pylori would not play a major role in the development of p53 or K-ras mutations. However, we cannot rule out the possibility that CagA(+) H. pylori is associated with the IL-1B polymorphism and p53 mutation.
